What mechanisms are being studied?

MK-677 is a non-peptide, orally active ghrelin receptor (GHS-R1a) agonist that stimulates natural pulsatile growth hormone release from the pituitary. Unlike injected GH, it maintains the body's natural feedback mechanisms. It increases GH amplitude without altering pulse frequency, elevates IGF-1 by 40-97%, and maintains these effects without tachyphylaxis over extended periods. It also significantly enhances deep (Stage IV) sleep quality.

What are the research questions?

• GH deficiency / age-related GH decline: 2-year study in elderly showed sustained IGF-1 increases to youthful levels without significant adverse effects.
• Muscle wasting / sarcopenia: Reversed diet-induced protein catabolism. Increased lean body mass in elderly subjects.
• Sleep quality: 50% increase in Stage IV (deep) sleep and 20% increase in REM sleep.
