Peptide Reconstitution Guide
Everything you need to properly prepare peptides — from supplies to step-by-step reconstitution and common mistakes.
Reconstitution Steps
Gather Materials
Peptide vial, bacteriostatic water (BAC water), U-100 insulin syringe (29–31 gauge), alcohol swabs. Work on a clean, flat surface.
Calculate Volume
Standard ratio: 1 mL BAC water per 5 mg peptide = 5,000 mcg/mL. For a 10 mg vial, use 2 mL for the same concentration. Write down your concentration for dosing calculations.
Swab Vial Tops
Use an alcohol swab on both the peptide vial and BAC water rubber stoppers. Wait 5–10 seconds for alcohol to evaporate before inserting needle.
Draw BAC Water
Insert syringe through BAC water stopper. Invert the vial and draw your calculated volume. Remove any air bubbles by tapping the syringe.
Add Water GENTLY
Aim the stream against the inside wall of the peptide vial — NEVER spray directly onto the lyophilized powder cake. Let the water trickle down slowly.
Let It Dissolve
Leave undisturbed for 1–2 minutes. The powder should dissolve on its own. If needed, gently swirl the vial. NEVER shake — shaking creates foam and shear forces that damage peptide bonds.
Store Immediately
Place reconstituted vial in refrigerator (2–8°C). The peptide is now ready to use. Typical shelf life: 2–4 weeks depending on the specific peptide.
Common Mistakes to Avoid
❌ Don't: Buy without verifiable COAs
Without third-party verification, you don't know what's in the vial. A COA should have an independent lab name, lot number, HPLC purity, and mass spec data.
❌ Don't: Start with 5 peptides at once
Start with one. Learn proper handling, observe responses, then add one at a time. This isolates variables and builds competence.
❌ Don't: Freeze reconstituted peptides
Ice crystals physically shear peptide chains. One freeze-thaw cycle can significantly reduce biological activity. Always refrigerate, never freeze.
❌ Don't: Skip the alcohol swab
Every unswabbed needle insertion risks contamination. Takes 3 seconds and protects your entire investment.
❌ Don't: Shake the vial
Creates foam and shear forces that denature peptide chains. Gently swirl or let it dissolve passively — never agitate.
❌ Don't: Use expired or cloudy peptide
Cloudiness = aggregation = degradation. Discard and use a fresh vial. Visual changes always indicate compromised product.
❌ Don't: Ignore the COA
2 minutes reviewing it protects your entire investment. Check HPLC purity (≥98%), mass spec match, lot number, and lab accreditation.
